No previous studies have investigated the use of graphite instead of activated charcoal in orchids. In this work, different concentrations of activated charcoal or graphite were added to KC medium to darken the culture medium and stimulate the in vitro propagation of Cattleya bicolor and of a double hybrid orchid (‘BLC Pastoral Innocence’). The seedlings were inoculated on growth regulator-free KC medium; the effects of activated charcoal (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) and graphite (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) concentrations were tested separately. Application of decolourized and partially purified polygalacturonase and α-amylase in apple juice clarification BJM

Polygalacturonase and α-amylase play vital role in fruit juice industry. In the present study, polygalacturonase was produced by Aspergillus awamori Nakazawa MTCC 6652 utilizing apple pomace and mosambi orange (Citrus sinensis var mosambi) peels as solid substrate whereas, α-amylase was produced from A. oryzae (IFO-30103) using wheat bran by solid state fermentation (SSF) process. These carbohydrases were decolourized and purified 8.6-fold, 34.8-fold and 3.5-fold, respectively by activated charcoal powder in a single step with 65.1%, 69.8% and 60% recoveries, respectively. Assessment of DNA extraction methods for detection of arbuscular mycorrhizal fungi in plant roots by nested-PCR Biological Sciences

DNA extraction methods were evaluated for the yield and purity of DNA recovered from mycorrhized roots and whether the recovered DNA is suitable for amplification of arbuscular mycorrhizal (AM) fungal SSU rDNA. The DNeasy Plant Mini Kit and three extraction buffers were used alone or in combination with either polyvinylpyrrolidone (PVP), polyvinylpolypyrrolidone (PVPP) and/or activated charcoal (AC). Among the extraction methods tested, those based on the CTAB buffers yielded more DNA than those based on the TE buffer and the DNeasy Plant Mini Kit. Cryopreservation of Cyrtopodium hatschbachii Pabst (Orchidaceae) immature seeds by encapsulation-dehydration Biocell

The aim of the present study was to investigate the efficiency of the encapsulation-dehydration technique for cryopreservation of Cyrtopodium hastchbachii Pabst seeds. Immature seeds of this species were cryopreserved by an encapsulation-dehydration technique. Seeds of five immature pods, 120 days after pollination, were encapsulated in 3% calcium alginate matrix and pretreated in liquid medium supplemented with 0.08 M sucrose (24 h), 0.15 M sucrose (24 h), 0.25 M sucrose (48 h), 0.5 M sucrose (24 h) and 0.75 M sucrose (24 h) in shaker at 60 rpm. Evaluation of nutrient supplementation to charcoal-treated and untreated rice straw hydrolysate for xylitol production by Candida guilliermondii BABT

Xylitol was produced by Candida guilliermondii from charcoal-treated and untreated rice straw hemicellulosic hydrolysate with or without nutrients (ammonium sulphate, calcium chloride, rice bran extract). Both, xylitol yield and volumetric productivity decreased significantly when the nutrients were added to treated and untreated hydrolysates. In the treated hydrolysate, the efficiency of xylose conversion to xylitol was 79% when the nutrients were omitted. The results demonstrated that rice straw hemicellulosic hydrolysate treated with activated charcoal was a cheap source of xylose and other nutrients for xylitol production by C. guilliermondii. Evaluation of oat hull hemicellulosic hydrolysate fermentability employing Pichia stipitis BABT

Oat hull hemicellulosic hydrolysate obtained by diluted acid hydrolysis was employed as fermentation medium for Pichia stipitis cultivation. A comparison between the use of treated hydrolysate with 1% activated charcoal to reduce the toxic compounds generated during the hydrolysis process and untreated hydrolysate as a control was conducted. In the cultures using treated hydrolysate the total consumption of glucose, low xylose consumption and ethanol and glycerol formation were observed. Aspasia variegata occurs naturally in the savanna of the Mato Grosso do Sul State, Brazil and it has been widely collected for its beautiful flowers. Additionally, its habitat has been greatly reduced and little or no investigation of its spread has been performed. In vitro shoot induction and multiplication from nodal segments of adult Ginkgo biloba plants Horticultura Brasileira

The in vitro performance of herbaceous and woody nodal segments from adult plants and the effect of hydrolyzed casein (HC 500 mg L-1), kinetin (KIN; 6-furfurylaminopurine 0.46 and 4.65 µM) and activated charcoal (AC 1.5 g L-1) were evaluated upon new shoots induction and development, and to establish a system of in vitro propagation from adult plants of Ginkgo biloba. Woody nodal segments did not produce axillary shoots and presented 100% of bacterial and fungal contamination in culture. Micropropagation of Alibertia edulis Rich BABT

The aim of this work was to develop a micropropagation protocol for Alibertia edulis, a medicinal Brazilian Cerrado plant. A. edulis seeds were sterilized and germinated in water-agar. Fifteen millimeter long explants from the seedlings cultivated for 50 days were used for the multiplication in three consecutive subcultures. The cytokinin benzyladenine (BA) alone or combined with indole-3-butyric acid (IBA) was added to ¼ MS medium. The treatments containing combinations of the BA + IBA induced more shoot proliferation than the BA alone, with the optimum concentration of 0.5 mg.L-1 for both growth regulators.
